Overview

Protein-protein interactions (PPIs) are the physical contacts established between two or more protein molecules, and they underlie virtually all cellular processes, from signal transduction and metabolism to gene regulation and structural organization. Mapping and understanding PPIs is central to molecular biology because most proteins function as part of complexes and networks rather than in isolation, and disruptions to these interactions are implicated in many diseases. The topic includes both experimental approaches—such as affinity purification coupled with mass spectrometry—and computational and bioinformatic methods used to predict, model, and analyze interactions and their networks. Related areas include proteomics, structural biology, interaction databases, and the use of interaction data in drug discovery and the interpretation of disease-associated genetic variants.
